Analysis of dinosaur bone cells confirms ancient protein preservation

ScienceDaily (Oct. 23, 2012) ? A team of researchers from North Carolina State University and the Palo Alto Research Center (PARC) has found more evidence for the preservation of ancient dinosaur proteins, including reactivity to antibodies that target specific proteins normally found in bone cells of vertebrates. These results further rule out sample contamination, and help solidify the case for preservation of cells -- and possibly DNA -- in ancient remains.

Dr. Mary Schweitzer, professor of marine, earth and atmospheric sciences with a joint appointment at the North Carolina Museum of Natural Sciences, first discovered what appeared to be preserved soft tissue in a 67-million-year-old Tyrannosaurus Rex in 2005. Subsequent research revealed similar preservation in an even older (about 80-million-year-old)Brachylophosaurus canadensis. In 2007 and again in 2009, Schweitzer and colleagues used chemical and molecular analyses to confirm that the fibrous material collected from the specimens was collagen.

Schweitzer's next step was to find out if the star-shaped cellular structures within the fibrous matrix were osteocytes, or bone cells. Using techniques including microscopy, histochemistry and mass spectrometry, Schweitzer demonstrates that these cellular structures react to specific antibodies, including one -- a protein known as PHEX -- that is found in the osteocytes of living birds. The findings appear online in Bone and were presented last week at the annual meeting of the Society of Vertebrate Paleontology.

"The PHEX finding is important because it helps to rule out sample contamination," Schweitzer says. "Some of the antibodies that we used will react to proteins found in other vertebrate cells, but none of the antibodies react to microbes, which supports our theory that these structures are surviving osteocytes. Additionally, the antibody to PHEX will only recognize and bind to one specific site only found in mature bone cells from birds. These antibodies don't react to other proteins or cells. Because so many other lines of evidence support the dinosaur/bird relationship, finding these proteins helps make the case that these structures are dinosaurian in origin."

Schweitzer and her team also tested for the presence of DNA within the cellular structures, using an antibody that only binds to the "backbone" of DNA. The antibody reacted to small amounts of material within the "cells" of both the T. rex and the B. canadensis. To rule out the presence of microbes, they used an antibody that binds histone proteins, which bind tightly to the DNA of everything except microbes, and got another positive result. They then ran two other histochemical stains which fluoresce when they attach to DNA molecules. Those tests were also positive. These data strongly suggest that the DNA is original, but without sequence data, it is impossible to confirm that the DNA is dinosaurian.

"The data thus far seem to support the theory that these structures can be preserved over time," Schweitzer says. "Hopefully these findings will give us greater insight into the processes of evolutionary change."

Dr. Marshall Bern, from PARC, performed the mass spectrometry. Former NC State doctoral student Timothy Cleland and research assistant Wenxia Zheng also contributed to the work, which was funded by grants from the National Science Foundation and the David and Lucile Packard Foundation.

    Video: Two-legged robot ?walks? a tightrope

    Researchers have modified a two-legged hobby-kit robot with the balance and dexterity required to inch its way along a thin?steel wire.?

    Japanese robotics researcher Masahiko Yamaguchi, who goes by the name Dr. Guero, modified the $1,800 hobby kit robot with grooves in its feet to catch the wire, akin to the way human tightrope walkers do with their toes, according to Gizmag.?

    The arms have fewer moving parts than the original to provide greater stability.

    Instead of walking ? that is putting one foot in front of the other ? the robot inches its way across the wire, which is about an eighth of an inch thick. Walking with alternating feet, Dr. Guero notes on his website, should be possible, but will require more work.

    No word on whether the robot will walk a wire between skyscrapers akin to Philippe Petit's 1974?stunt between the World Trade Center towers featured in the Academy Award winning "Man on Wire," but given the robot's nerves-of-steel such a feat should be as easy as cruising over table inside a lab.

    Effect of the Hispanic vote in Vegas may not stay in Vegas

    LAS VEGAS?This city has seen more political commercials than anywhere else in 2012, more than anywhere else in the history of presidential campaigns: 73,000 TV ads, according to The New York Times. In an election with only a handful of states that are considered up for grabs, the importance of Nevada's six electoral votes was highlighted when President Barack Obama chose Lake Las Vegas, a golf resort east of here, as the place to spend three days preparing for the first presidential debate.

    The state's Hispanic voters are especially sought-after: According to Jim Messina, the Obama campaign manager, Latino voters are particularly crucial this year in Nevada, as well as in Colorado and Florida.

    "As evidenced by the explosive growth among Latinos, Nevada offers a case study in the demographic changes that are transforming large swaths of the country and, in the process, reshaping the electoral map," David Damore, a political scientist at the University of Nevada, Las Vegas, told Yahoo News. "As older white voters die out, they'll be replaced by nonwhite voters."

    There are more than?50.5 million Hispanics in the United States. That's a 43 percent increase in the Hispanic population in the past 12 years.

    Yahoo News headed to Nevada earlier this year to find out what's on the minds of some of the most important voters in one of the most important states of Campaign 2012.

    'I don't want to vote for the wrong guy. But it's really hard to find out who the wrong guy is.'

    Austreberto Hernandez, known as "Asti," will be a junior at the University of Nevada, Las Vegas this fall, studying political science and business management. This summer he worked two jobs, registering voters for Mi Familia Vota and working as an intern for the?Latin Chamber of Commerce Community Foundation.

    Hernandez and his six siblings were all born in the United States: Their father came across the border from Mexico looking for work?in the 1980s and became a citizen because of the?Immigration Reform and Control Act signed by Ronald Reagan in 1986.

    "Ever since I can remember we would go from city to city looking for work," Hernandez, a registered independent, told Yahoo News. "Vegas was booming in the early '90s. It was just exploding. It was blowing out of the water. So everyone just headed here."

    'I'm a Hispanic Republican. We're almost a minority inside a minority.'

    Omero Alexandro Garza, who goes by "Alex," started working as a child in his family's truck stop. He taught middle school and then worked in real estate, starting out part-time at Century 21, then eventually founding his own real estate firm before moving on to other jobs in the industry.

    "Without question, the primary issue for Hispanics is the economy," says Garza. "Immigration is near and dear to every Hispanic's heart. It's always going to be important to us. But more important is whether or not we have a job."

    Garza's father came to the United States from Mexico as an undocumented immigrant. "He gained his amnesty under Ronald Reagan so it kind of just stems from there," Garza told Yahoo News, referring to his status as a registered Republican.

    His house is underwater. He says he put down $80,000 on a $400,000 mortgage for a home that is now valued at $190,000. His wife has taken a job teaching English in Abu Dhabi, so the family is in the midst of moving to another country. "I mean I don't want to cry, but it's difficult because this was our hopes and dreams; this is where we raised our kids," Garza says. "It's the only house my children have known. It's difficult to go from stability within your family to instability and uncertainty."

    He plans to vote by absentee ballot for Mitt Romney.

    'If you look at the demographics, we are going to be the majority.'

    Helena Garcia, 53,?styles herself "La Protectora." She's a real estate broker who also co-founded Latinos in Action, a pro bono advocacy group that tries to help people who think they have been cheated.

    "If I only had to do real estate I would be very unhappy," Garcia told Yahoo News, adding that she prefers "going out into the community and seeing people's problems and devastation."

    Garcia's family came to Las Vegas in 1956 from Juarez, Mexico. Her father worked as a dishwasher at a Mexican restaurant. Although she's a registered independent, she sides with the Democrats.

    "In my opinion, most of the people in the Republican Party are racist and the majority of them want that more conservative view of 'Go back to where you came from,'" Garcia says. "There is no excuse not to pass the DREAM Act when these kids are going to better our country; they are going to bring more education to our country. Most of them are fully bilingual so you bring more culture and languages to our country. There is no other reason not to pass the DREAM Act except for racism."

    Video conferencing startup Blue Jeans Network is all about making video conferencing easier and more accessible for companies, with a cloud-based way to connect video conferences between existing infrastructure and even consumer-facing apps like Skype. Today it?s giving its users one more way to connect ? with LinkedIn.

    By integrating with LinkedIn, Blue Jeans will allow users to use their social network credentials to sign in. Once they?ve connected, other users they are video conferencing with will be able to see their headshot, bio, and other information stored on LinkedIn, as it will be automatically uploaded to their Blue Jeans profile. That will give members the ability to learn more about people on the call, without having to click through to other sites. They?ll also be able to send each other messages offline through LinkedIn during and after the conference.

    For Blue Jeans, the LinkedIn integration will offer up more ways to connect, particularly for those in the HR and recruiting field. One advantage Blue Jeans has over other conferencing solutions is that users don?t have to ?buddy up? to take calls with one another. Recruiters can create temporal meetings, without having to worry about being contacted on Skype by interested applicants who aren?t quite right for a particular job. LinkedIn has more than 175 million members across 200 countries, so it offers a pretty large user base who can take advantage of the feature.

    The integration follows on previous work that Blue Jeans had done to allow users to connect with their Facebook credentials. It also announced in September an integration with Salesforce.com that will allow users to make calls from directly within the Salesforce app.

    Blue Jeans has raised about $50 million since being founded in 2009. That includes a $25 million round of financing from NEA, Accel, and Norwest Venture Partners that it raised earlier this summer. The company is on pace to enable 25-35 million minutes of video in a year, which Stu Aaron, chief commercial officer of the startup, estimates is about 10 percent of the enterprise video conferencing market.


    At Blue Jeans Network, their mission is to make video communications as easy and pervasive as audio communications, enabling more effective collaboration at work, at home, and on the road. Their cloud-based conferencing service enables people to connect with each other any time, any place, and from practically any device.

    'Impartial expert' behind new Web copyright rules lobbied for music ...

    Internet users will soon be subject to a new ?six strikes? system that'll let ISPs cut-off customers caught sharing files illegally. And while the program has received the blessing of the major labels, the music biz may have more than a bit part.

    Four years after Internet Service Providers first considered implementing a new system to deter copyright law violations, the largest telecommunication companies in the United States are expected to finally put into place a ?six strikes? program later this year that will punish repeat offenders with sanctions of increasing severity after each instance of illegal file sharing is identified.

    The program is largely the brainchild of the Center for Copyright Information, who last year said, "We anticipate that very few subscribers, after having received multiple alerts, will persist (or allow others to persist) in the content theft.? Through the six-strikes program expected to be installed before 2013, customers caught using peer-to-peer programs to illegally share copyrighted files will be subjected to warnings, legal notices and even have their connection speed throttled.

    Recently the CCI acknowledged that an ?independent and impartial technical expert? was brought on board to review the ?accuracy and security? of the technology that is soon expected to be adopted by AT&T, Cablevision Systems, Comcast, Time Warner Cable and Verizon, but further investigation indicates that they did anything but.

    A post published this week to the CCI?s official blog on copyrightinformation.org acknowledges that ?recognized technology expert? Stroz Friedberg will be put in charge of evaluating the ?MarkMonitor? system that will be used ?for identifying alleged infringement over peer-to-peer networks.? And while Friedberg will indeed bring with him to the CCI a career in copyright law that might be unmatched, he also takes with him a cozy relationship with the music biz which reveals anything but an impartial alliance.

    Cory Doctorow authored another post on BoingBoing.net this weekend that shines a light on Mr. Friedberg, and the checkered past he?s has with some very cozy comrades of the CCI: the Recording Industry Association of America.

    ?Stroz Friedberg has completed its initial review of MarkMonitor?s methodologies and found that the system is accurate and works properly,? the CCI writes. Nowhere, however, does it acknowledge that Friedberg lobbied extensively on behalf of the RIAA going back to 2004, who retained him at a cost of more than half-a-million-dollars over the course of five years.

    The RIAA, the largest trade group of its kind representing major America record labels, made it well known in Hollywood and Washington alike that they will not stand for copyright infringement, lobbying endlessly in attempt to have stringent Internet laws passed to put them in control of how content is distributed and to whom. It?s no surprise that the RIAA has endorsed the ?six-strikes? program, which can effectively cut off connections for repeat offenders, but the back-handed hiring of one of their own lackeys comes to a surprise, even in an age when dirty tricks seems all the norm.

    ?It?s a disappointing choice, particularly in light of CCI?s professed desire to build public confidence in CAS and the fairness of its processes. It would have been refreshing to see an academic computer scientist or some other truly independent party appointed to fill that important role,? University of Idaho Law Professor Annemarie Bridy tells TorrentFreak.

    According to the Center for Responsive Politics, Friedberg has most recently lobbied for the RIAA to the tune of $15,000 in 2009, raking in a total of $637,000 since coming on board five years earlier.

    ?CCI?s choice of a former RIAA lobbying firm makes it clear that the copyright owner parties to the Memorandum of Understanding were more interested in appointing someone they trust than in appointing someone the public can trust,? Birdy adds.

    The CCI?s six-strike program has also been endorsed by the White House, who previously remarked that the system should ?have a significant impact on reducing online piracy.? The Motion Picture Association of America (MPAA), the trade-group that does for the silver screen what the RIAA does for the record industry, has also thrown their weight behind the six-strike program. Coincidently, they also have some major clout in Washington: the MPAA is headed by former Senator Chris Dodd, a close pal of Vice President Joe Biden.

    White House orders spy agencies to share cyberthreat intel with companies

    WASHINGTON (AP) -?A new White House executive order would direct U.S. spy agencies to share the latest intelligence about cyberthreats with companies operating electric grids, water plants, railroads and other vital industries to help protect them from electronic attacks, according to a copy obtained by The Associated Press.?

    The seven-page draft order, which is being finalized, takes shape as the Obama administration expresses growing concern that Iran could be the first country to use cyberterrorism against the United States. The military is ready to retaliate if the U.S. is hit by cyberweapons, Defense Secretary Leon Panetta said. But the U.S. also is poorly prepared to prevent such an attack, which could damage or knock out critical services that are part of everyday life.?

    The White House declined to say when the president will sign the order.?

    The draft order would put the Department of Homeland Security in charge of organizing an information-sharing network that rapidly distributes sanitized summaries of top-secret intelligence reports about known cyberthreats that identify a specific target. With these warnings, known as tear lines, the owners and operators of essential U.S. businesses would be better able to block potential attackers from gaining access to their computer systems.?

    An organized, broad-based approach for sharing cyberthreat information gathered by the government is widely viewed as essential for any plan to protect U.S. computer networks from foreign nations, terrorist groups and hackers. Existing efforts to exchange information are narrowly focused on specific industries, such as the finance sector, and have had varying degrees of success.?

    Yet the order has generated stiff opposition from Republicans on Capitol Hill who view it as a unilateral move that bypasses the legislative authority held by Congress.?

    Administration officials said the order became necessary after Congress failed this summer to pass cybersecurity legislation, leaving critical infrastructure companies vulnerable to a serious and growing threat. Conflicting bills passed separately by the House and Senate included information-sharing provisions. But efforts to get a final measure through both chambers collapsed over the GOP's concerns that the Senate bill would expand the federal government's regulatory power and increase costs for businesses.?

    The White House has acknowledged that an order from the president, while legally binding, is not enough. Legislation is needed to make other changes to improve the country's digital defenses. An executive order, for example, cannot offer a company protection from liabilities that might result from a cyberattack on its systems.?

    The addition of the information-sharing provisions is the most significant change to an earlier draft of the order completed in late August. The new draft, which is not dated, retains a section that requires Homeland Security to identify the vital systems that, if hit by cyberattack, could "reasonably result in a debilitating impact" on national and economic security. Other sections establish a program to encourage companies to adopt voluntary security standards and direct federal agencies to determine whether existing cyber security regulations are adequate.?

    The draft order directs the department to work with the Pentagon, the National Security Agency, the director of national intelligence and the Justice Department to quickly establish the information-sharing mechanism. Selected employees at critical infrastructure companies would receive security clearances allowing them to receive the information, according to the document. Federal agencies would be required to assess whether the order raises any privacy or civil liberties risks.?

    To foster a two-way exchange of information, the government would ask businesses to tell the government about cyberthreats or cyberattacks. There would be no requirement to do so.?

    The NSA has been sharing cyberthreat information on a limited basis with companies that conduct business with the Defense Department. These companies work with sensitive data about weapon systems and technologies and are frequently the targets of cyberspying.?

    But the loss of valuable information has been eclipsed by fears that an enemy with the proper know-how could cause havoc by sending the computers controlling critical infrastructure systems incorrect commands or infecting them with malicious software. Potential nightmare scenarios include high-speed trains being put on collision courses, blackouts that last days or perhaps even weeks or chemical plants that inadvertently release deadly gases.?

    Panetta underscored the looming dangers during a speech last week in New York by pointing to the Shamoon virus that destroyed thousands of computer systems owned by Persian Gulf oil and gas companies. Shamoon, which spreads quickly through networked computers and ultimately wipes out files by overwriting them, hit the Saudi Arabian state oil company Aramco and Qatari natural gas producer RasGas.?

    Panetta did not directly connect Iran to the Aramco and RasGas attacks. But U.S. officials believe hackers based in Iran were behind them.?

    Shamoon replaced files at Aramco with the image of a burning U.S. flag and rendered more than 30,000 computers useless, Panetta said. The attack on RasGas was similar, he said.?

    A spokeswoman for the National Security Council, Caitlin Hayden, said the administration is consulting with members of Congress and the private sector as the order is being drafted. But she provided no information on when an order would be signed. "Given the gravity of the threats we face in cyberspace, we want to get this right in addition to getting it done swiftly," she said.

    Latest LG Nexus 4 leak provides minor new details

    It's been a while since we've had any fresh LG Nexus 4 leaks, largely because much of the upcoming phone's hardware has already been laid bare. Nevertheless, notorious Twitter leaker @evleaks has today revealed a few minor, yet previously unknown specs, along with a new photo of the device's front face. The Twitter post also confirms earlier leaked specs, including a quad-core Snapdragon S4 Pro CPU, 2GB of RAM, and 8GB and 16GB storage options.

    According to the latest leak, the Nexus 4 will be slightly thicker than its predecessor, measuring 9.1mm -- though let's remember that there's no unsightly hump down below this time around. In addition, weight is reported as 139 grams, a little heavier than the GSM Gnex's 135 grams. And the internal, non-replaceable battery is said to be rated at an ample 2100mAh.

    Capacity is one thing, but it'll be interesting to see if the Nexus 4 sports the same "800 charging cycles" tech that LG's been pimping with the Optimus G as an increase over the competition. We're also curious to see whether, like newer Nexus 7 models, the Nexus 4 hardware supports the new power management features of the 3.4 Linux kernel.

    On software, evleaks reports Android "JellyBeanMR1" as being loaded onto the device -- that's the same numberless version we've seen floating around on other prototypes. It's widely expected that the final device will ship with Android version 4.2, which may or may not wear the Jelly Bean badge.

    So the bottom line is this -- slightly thicker, a tiny bit heavier, and you shouldn't need to worry too much about battery life. Of course, we'll reserve final judgment until we get our hands on the finished product, likely at Google's "Playground" event on Oct. 29.
